If I run Discover Roles again, do I overwrite role assignments already made?

No. When you run Discover Roles again, you overwrite the existing list of device role recommendations that were made by the previous discover roles operation, but for which the role has not been assigned. Role discovery has no effect on devices that have an assigned role.
